Normal lymphoid cells have a low side scatter and are CD45++. Haematogones, which are immature lymphoid progenitors, are CD45 dim and tend to have a lower side scatter. They are usually evident as distinct populations in the the bone marrow, especially in paediatric patients.

Flow Cytometry: Lymphocyte Gate
On the side scatter vs CD45 plot, lymphocytes are SSc-low/CD45++. Haematogones are immature lymphoid precursors, and have a very low side scatter and dim CD45.

Cell populations in the lymphocyte window:

  • Normal B cells
  • Normal T cells
  • Normal NK cells
  • Some contaminating monocytes (there is some overlap between the monocyte gate and the lymphocyte gate)
  • Abnormal lymphoid cells (of all lineages, but especially indolent B lymphoid neoplasms)
  • T large granular lymphocyte leukaemia
